The 56 as a amplification factor a bit higher than the 27 (14 instead of 9). If I replace one 27 by one 56, I will see a difference in amplification? Amplification will be higher in my audio power amp or not really?

Depends on the circuit. As a 1st audio amplifier tube you will have more gain. Too high of gain can cause a radio to go into oscillation. As an oscillator or diode connected detector there will be no difference in gain.
Don't try interchanging these tubes if several are biased together, such as in Sparton radios. Grid cutoff is different when tubes have higher gain.
