I just recapped all parts one tube 6b5 doesn't light up and I hear a hum sound in speaker what do I do next

With an ohmmeter measure the resistance between pins 1 and 6, the two big pins. If there is infinite resistance, the filament is open. If the resistance is open, may try heating and resoldering the two pins in case there is a bad solder connection.
